Mother Road 66 is an area located in the Midwest in the United States referencing the historic road U.S. Route 66 which passes through it.

Mother Road 66 is the nickname given for the famous U.S. Route 66 that stretches from Chicago, Illinois (pictured) to Santa Monica, near Los Angeles, California. The road covers a total of 2,448 miles (3,940 kilometres) and goes through the states of Illinois, Missouri, Kansas, Oklahoma, Texas, New Mexico, Arizona and California.

Mother Road 66 is a continuous road that goes through the various states in the US, and begins in the lower sections of Chicago in the game, moving south towards St. Louis and later nearby Amarillo. The road is guided by recurring appearances of the Route 66 sign on the side of the road which can guide the players through this vast trail, though this signage abruptly stops near the Central Woods area in-game, which requires players to continue on their own path along this road.

The road stretches along various landmarks that are present along Route 66, and captures the varying landscape of this traversing road through woods and deserts. The road becomes unclear after Barstow located south of Mojave, marking this section to supposedly be the end of the road, though most individuals consider the road to traverse through Palm Springs and unofficially end at the Santa Monica pier located in Los Angeles, as in-game, various Route 66 emblems and signs can be seen on the Santa Monica Pier.

The road demonstrates the essence of driving through the bypassed road in-game with many of the original key points and locations that were captured along the road during its peak time of use. Many abandoned buildings can be seen when driving along the side of the road with old representations of filling stations and buildings. Another common sight to see on Route 66 is the vast array of motels and eateries that are present when driving, typically located near towns. References to real life such as the Wigwam Motel (Holbrook) or Roy's Motel and Café, which has been converted to "Jay’s Hotel Eatery” in-game, located in the abandoned small ghost town named Amboy.

There are numerous fictional motels and stops with names such as "Route 66 Motel" amongst others. Other restaurants such as the Twin Arrows Trading Post being renamed as "Twins Restaurant” in Arizona, with an easter egg encompassing the bypass of Interstate-40, with quote “Best Little Stop on I-40” being written on the side of the abandoned restaurant. Interstate-40 resemblances can also be seen south of the Mother Road, with a 4 lane highway stretching from origins near Los Angeles moving past Dallas near Cape Canaveral. Towns such as Holbrook, Winslow, Barstow, Kingman and Amboy are located along the route with individual references to the game.
